The Joker is very much obsessed with the Wayne family. Why? Perhaps it's because he is one of them. This is based on the idea that he is the result of Thomas Wayne and Mrs. Fleck's affair in the past.
It doesn't take a genius to figure out how much Arthur loves his mom. It seems that his relationship with her is going to be a pivotal force in his life. And who knows, her mother's death could be the reason he becomes the villain we all know.
How did the Joker's mom fall mentally ill? Well, Thomas Wayne could have been the reason. This theory is based on the idea that he and Mrs. Fleck had a romantic connect and that it didn't end well for the Joker's mother.
The trailer opens with a few shots of Arthur Fleck AKA Joker, who seems to be struggling with depression. He is seen talking to a social worker about his day and then walks down through Gotham city in a slouched posture.
We also see shots of Arthur bathing his mother and dancing with her later, which only suggests that his mother's mental health is obviously not in its best state.
When his mother dies, his motivation as the Joker is to seek vengeance on people who might be responsible for it, specifically Wayne Enterprises. Perhaps because her mother's medication is too high a price for him to afford it.
If you watch Joker's trailer closely, you would see that a lot of imagery in the clip represents the division between the privileged and the underprivileged. You see those men clad in a suit? They obviously represent the upper-class.
The mob of people wearing clown masks could have been influenced by Joker to be in revolt against those in power. The train incident could have been Joker's last straw or boiling point.
